牡丹R2R3-MYB转录因子PoMYB96的克隆及对多重非生物胁迫的响应

    目的 高温、低温、干旱和高盐等多种非生物胁迫严重影响牡丹的正常生长,限制了牡丹的生产栽培。大多数MYB转录因子在植物抵抗逆境胁迫的过程中起到正向调节作用,保障了植物正常的生长发育。克隆并探究牡丹PoMYB96基因在非生物胁迫下的表达模式,可为深入研究牡丹抵抗非生物胁迫提供理论参考。
    方法 本研究采用RT-PCR技术克隆‘凤丹’(Paeonia ostii ‘Feng Dan’)牡丹的PoMYB96基因并对其序列信息及所编码蛋白的特征进行系统分析。牡丹PoMYB96基因与拟南芥(Arabidopsis thaliana)MYB家族成员进行系统进化分析并构建进化树,采用实时荧光定量PCR技术对PoMYB96基因在牡丹叶片中响应多种非生物胁迫过程中的表达模式进行分析。
    结果 PoMYB96基因的cDNA序列全长为984bp,编码区氨基酸数量为327个;PoMYB96蛋白存在两个SANT保守结构域;PoMYB96与拟南芥AtMYB96同源性最高;PoMYB96基因启动子含有厌氧、水杨酸和光响应元件以及ABA、赤霉素、茉莉酸等激素响应元件;PoMYB96在牡丹的根、茎、叶、花、雄蕊、雌蕊中都有表达,在叶片中的表达水平最高;在低温、高温、干旱、水淹和盐胁迫下的相对表达量均上调。
    结论 PoMYB96基因可能参与牡丹对多种非生物胁迫的响应,这为研究牡丹抵抗非生物胁迫提供了理论参考,为利用分子育种改良牡丹抗性提供了候选基因。

     

    Abstract:
    Objective PoMYB96 related to the response to abiotic stresses of Paeonia ostii was cloned to analyze the roles it played in regulating growth of the plant.
    Method The R2R3-MYB transcription factor PoMYB96 that regulates the response of tree peony (P. ostii Fengdan) to high or low temperature, drought, and salinity stresses, which can severely hinder the normal growth of the plant, was cloned using RT-PCR. Sequence and encoded protein of the gene were analyzed. An evolutionary tree was constructed between PoMYB96 and MYB family members from Arabidopsis thaliana. Expressions of the gene in the leaves under high and low temperatures, draught, flooding, and high salinity were determined by qRT-PCR.
    Result The full-length cDNA sequence of PoMYB96 was 984bp encoding 327 amino acids. The protein contained two conserved SANT domains and shared the highest homology with AtMYB96 of A. thaliana. Its promoter region harbored anaerobic-, salicylic acid-, and light-responsive elements, as well as hormone-responsive elements for ABA, gibberellin, and jasmonic acid. The gene was expressed in the roots, stems, leaves, flowers, stamens, and pistils, with the highest level observed in the leaves. Its relative expression was significantly upregulated under low or high temperature, drought, waterlogging, and salt stress.
    Conclusion PoMYB96 participated in the response of tree peony to abiotic stresses and could be a candidate marker for molecular breeding selection.
