药用菊花花型性状与ISSR和SCoT标记关联分析

ISSR and SCoT Molecular Markers Related to Floral Phenotypes of Chrysanthemum morifolium

    目的 筛选与药用菊花花型性状紧密关联的分子标记,为分子标记辅助育种提供技术依据。
    方法 本研究选用11条简单序列重复区间扩增(inter-simple sequence repeat, ISSR)及20条起始密码子靶向多态性(start codon targeted, SCoT)标记,对64份药用菊花种质开展全基因组多位点扫描;运用OriginPro 2024和Structure 2.3软件分析其表型遗传多样性和分子群体结构;借助Tassel 5.0软件对9个花型性状与分子标记开展关联分析。
    结果 64份药用菊花种质的9个花型性状的变异系数为30.67%~82.72%;31条标记在64份种质中累计扩增出197个多态性位点,多态性比率平均为88.33%;多态性信息量值(polymorphism information content, PIC)为0.57~0.87,平均值为0.78;相关性分析表明,花型性状间存在显著或极显著正相关;花型性状聚类分析表明,64份药用菊花种质可聚为大花型、小花型和中间型3大类;当K=4时,ΔK值达最大值,64份种质被划分为4个类群,其中Q>0.6的种质占比81.25%;关联分析发现,12个标记位点与8个花型性状显著关联(P<0.01),各位点对花型变异的解释率为11.74%~29.27%,其中ISSR37对外舌状花长的解释率最高。
    结论 药用菊花花型性状遗传多样性丰富,所获得的12个与花型显著关联的分子标记可为高产、优质菊花新品种选育提供分子辅助手段。

     

    Abstract:
    Objective Molecular markers closely related to the floral phenotypes of Chrysanthemum morifolium were identified to facilitate breeding selection.
    Methods Based on 11 ISSR and 20 SCoT markers, 64 accessions of medicinal chrysanthemum (C. morifolium) were scanned for multiple genomic loci. On selected 9 floral phenotype traits, genetic diversity and population structure were analyzed using OriginPro 2024 and Structure 2.3 software. Association of the traits with the ISSR/SCoT markers was examined with Tassel 5.0.
    Results The coefficients of variation on the 9 traits ranged from 30.67% to 82.72%. The markers generated 197 polymorphic loci with polymorphic information contents between 0.57 and 0.87, averaging 0.78. Significant or extremely significant correlation was observed between most of the traits. The 64 accessions were clustered into three primary classes by flower size, i.e., large, small, and intermediate. And, at the optimal grouping at K=4 (ΔKmax) for classifying population structure, they were divided into 4 subpopulations that covered 81.25% of all with a membership probability (Q) greater than 0.6. Out of the 31 markers, 12 loci significantly correlated with 8 flower traits (P<0.01) explaining 11.74-29.27% of the phenotypic variations. And the association between ISSR37 and the length of ray floret was most significant.
    Conclusion The collected 64 C. morifolium germplasms were genetically rich in diversity on the floral phenotypes but relatively simple in relationship. The 12 trait-related ISSR/SCoT markers identified in this study could be of value for breeding selection.
