石刁柏体细胞胚的培养与植株再生研究

摘要: 石刁柏的外植体在附加激素的MS固体培养基上诱导愈伤组织。在愈伤组织的分类选择中获得的胚性愈伤组织,转移到含2,4-D1～1.5mg/L的液体培养基中悬浮培养繁殖。在继代培养中,将颗粒大于1mm的愈伤组织,转移到无激素的MS培养基中诱导体细胞胚发生,提高培养基中琼脂的浓度,可减轻体细胞胚的玻璃化。非玻璃化的棒状胚在1/2MS固体培养基上发芽获得了完整植株。

Abstract: Callus of Asparagus officinalis L.was induced on MS solid medium with bormone Embryonic callus was obtained from the selection and classification of the callus.Embryonic callus was proliferated by suspension culture in contained2,4-D 1～1.5mg/L MS liquid mediurn,Callus of more than lmm was inoculated on hornlone-free MS medium to induce somatic embryos.As agar concentration was increased from 0.6%to 0.8% or 1.0%,vitrification of somatic embryos decreased.Non-vitrified clubshapped embryos gern1inate and develop into normal plants on 1/2MS hormone-free solid medium.
