提高龙眼离体培养成活率的研究
Studies on Improving Survival Rate of Longan (Dimocarpuslongan Lour.) in Vitro Culture

摘要: 表面污染和材料褐变是龙眼组织离体培养成活率低的主要原因,采用切取中间茎段内部组织培养和多种药液交替浸泡的前处理方法,是降低污染率、褐化率,成功建立无菌初代培养物的有效措施;不同物候期的茎段,诱芽率不同,以夏、秋季的茎段较为适合作为起始外植体;选用MS基本培养基诱芽,诱芽率可达73%左右,且生长整齐,叶色较绿,为以脱毒为目的的龙眼茎尖培养研究提供无菌、无褐变的茎尖。Abstract: Microbial contamination and browning were the main reason causing the low survival rate of longan tissue culture in vitro.Using the internal tissue of middle stem sections and alternately immersed with multiple disinfectants as pretreatment can decrease contamination and browning.The stem germination numbers varied with seasons, and the stems in summer or autumn was suitable as explants.The rate of responsive explants could reach 73% when they were culture on MS media, and the adventitious stem grew very well.
