惠阳胡须鸡LPL、FASN基因多态性的研究

Restriction Fragment Length Polymorphism of LPL and FASN Genes in Huiyang Bearded Chicken

  • 摘要: 以惠阳胡须鸡为研究对象,分别以LPL、FASN基因为脂肪性状相关的候选基因,采用限制片段长度多态性(RFLP)检测这2个候选基因在惠阳胡须鸡中的多态性。研究结果表明,(1)用限制性内切酶HinfⅠ对LPL基因第8内含子进行RFLP分析,发现76.3%为已报道的BB型,另发现了两种新的基因型,分别占1.7%和22%,无AA型;(2)用限制性内切酶HaeⅢ对FASN基因进行RFLP分析,发现BB基因型比例为64.8%,BC型12.24%,CC型基因型比例23.4%。B等位基因频率为0.7041,C等位基因频率为0.2959,B等位基因频率明显高于C等位基因频率。

     

    Abstract: Restriction fragment length polymorphism (RFLP) of LPL and fatty acid synthetase (FASN) genes in Huiyang bearded chickens was obtained to facilitate the selection for dominant genotypes associated with the high-quality meat and fast-growing characteristics in chicken. Genomic DNA was extracted from the chicken blood. The two genes of interest were amplified by PCR. The amplied product of LPL gene was digested by Hinf I,and FASN gene by Hae Ⅲ, for RFLP analysis. The results showed that (1) in LPL gene,the genotype BB was 76.3%,while the two newly discovered genotypes were 1.7% and 22%, with no AA type detected; (2)in FASN gene, genotype BB, CC, and BC were 64.8%, 23.4%, and 12.24%, respectively,while the frequencies of B and C genes were 0.7041 and 0.2959, respectively.
