Identification and Phylogenetic Analysis of Bacillus Isolated from Soil at 1st Pit Site of Emperor Qin's Terracotta Warriors

Abstract:
Bacillus bacteria are widely distributed around the world.This study examined the diversity of Bacillus in the pit soil at Terracotta Warriors site and the geographical distribution of the bacteria in the specific environment.Soil samples were collected from the 1st pit of the Terracotta Warriors and subjected to a temperature screening and dilution plating to obtain 10 strains of Bacillus.These strains were subsequently identified by their 16S rRNAs to belong to 4 species and two possibly new Bacillus species.The 16S rRNA similarity among the strains was more than 99%.Thus, abundant Bacillus species were found to exist in the soil of the Terracotta Warriors site, and that the geographic distribution enriched the national microbial resource.
Lactobacillus Population and Properties of Soy Yoghurt in Cold Storage

Abstract:
Using soybean as the protein basis, yoghurt was made by inoculating FJAT-7928(Streptococcus thermophilus)and FJAT-7929(Lactobacillus bulgaricus).The lactic acid bacteria population, acidity and pH of the yoghurt stored at 4℃ were determined.The result showed that the soy yoghurt prepared by the co-fermentation had a maximum number of the bacteria of 3.5×109 cfu·mL-1, which was more than those in the soy yoghurt inoculated with either FJAT-7928 or FJAT-7929 alone.After 30 days of the cold storage, the acidity of the co-fermented yoghurt was 109.0°T, which was in between 114.0°T for FJAT-7929 yoghurt and 60.0°T for FJAT-7928 yoghurt.The final pH of the co-fermented yoghurt was 3.58, which was lower than 3.65 for FJAT-7929 yoghurt or 4.09 for FJAT-7928 yoghurt.At 4℃, the Lactobacillus population, acidity and pH of all three yoghurt products remained constant after 30 days in storage.
Growth Factors, bFGF and IFG-1, in Pectoral Fin Passage Cells of Anguilla anguilla

Abstract:
To establish a model for cell isolation and identification, as well as to gain an in-depth understanding of the viral diseases in Anguilla anguilla, the roles played by the growth factors, bFGF and IFG-1, on the proliferation of the eel's pectoral fin cells were studied.Under in vitro conditions, the cells were collected, counted and growth curves constructed.On the L-15 medium containing 10% FBS, bFGF showed a significant evidence in promoting the growth of the pectoral fin cells.The cellular growth peaked at bFGF concentration of 10 μg·L-1.On the other hand, IGF-1 in the concentrations from 5 μg·L-1 to 50 μg·L-1 did not show any apparent effect on the growth.Therefore, a culture medium formulation for effective proliferation of the A.anguilla pectoral fin cells was obtained.This would, in turn, aid future studies on the pathogen's isolation, identification and vaccine preparation.
Biological Functions of Mah-TrxA Fusion Protein from Aeromonas hydrophila

Abstract:
This experimentation was designed to help understand the antigenic and immunogenic rules of Aeromonas hydrophila (Ah). Indirect enzyme-linked immunosorbent assay showed the ratios of the antibody titers of the rabbit anti-ZN1-outer membrane protein (OMP) serum to the expression product, Mah-TrxA, and the recombinant protein of thioredoxin and a major adhesion agent and ZN1-OMP to be 1∶3 200 and 1∶12 800, respectively; and the ratios of the antibody titers of the rabbit anti- Mah-TrxA serum to Mah-TrxA and ZN1-OMP to be 1∶12 800 and 1∶1 600, respectively.Carp epithelial tumor cells (EPC) were used to study the adhesion effect of different sources and serotypes of Ah to EPC, as well as the biological functions played by Mah-TrxA.The adhesion test indicated that Ah were separated from the fish, the serotypes were O∶Ah10501, O∶9, and O∶CQ-1; the adhesion rate of Ah to EPC was greater than 97%; ATCC7966 was separated from milk, the serotype was O∶1; and, the adhesion rate of ATCC7966 to EPC was 78.5%.The results indicated that Mah-TrxA and the rabbit anti-Mah-TrxA serum could significantly inhibit or block the bacterial adhesion to EPC.It suggested that the fusion protein expressed through genetic engineering techniques in Ah had similar antigenicity and immunogenicity as the wild strain, ZN1.
The Study of Correlation of Three Different Avian Paramyxovirus Antigens

Abstract:
The antigenic relationship of three Avian Paramyxovirus,Goose paramyxovirus (MQG strain), duck paramyxovirus (Lg strain) and Newcastle disease virus (F48E8 strain) were determined by using virus cross-neutralization tests(NT) and cross-hemagglutination inhibition test (HI). The results indicated that the R value among three viruses was lower than 0.2 and revealed that there were clear antigenic variations among these Avian Paramyxovirus.
Breeding on the Two New Meat Strains of Liancheng White Duck

Abstract:
Two new meat strains of Liancheng white duck (I and II) were selected from Lijia duck and Liancheng white duck as parents by the methods of the technology of hybridization and a multi-generation transversely breed, and colony continuing selection, and the traits of white feather, beak and green feet were fixed gradually. After 3 and 4 generations dreeding, the compositive trait rate of white feather, black beak and green feet in the two new meat strains was improved from 0% to 99.01% and 99.35%, and mean heritability of 0.808 9 and 0.951 7 was achieved, respectively. At the age of 7 weeks, the average bodyweight and the average feed conversion efficiency of the male and female ducks in strainⅠand strain II was 1824.64±170.90 g and 2.93∶1, 2168.22±219 g and 2.78∶1,respectively.Compared with the basic population-Liancheng white duck, the growth rate of the two new strains was increased by 95.0% and 131.6% and the feed conversion efficiency by 12.3% and 16.8%, respectively. The results showed that the selection effects were extremely significant.
HPLC-MS/MS Determination of Clonidine Hydrochloride and Cyproheptadine Hydrochloride Contents in Pig Urine

Abstract:
A methodology applying HPLC-MS/MS was developed for the determination of clonidine hydrochloride and cyproheptadine hydrochloride contents in pig urine.Samples were hydrolyzed with perchloric acid, extracted with ethyl acetate-isopropanol, and purified through an MCX SPE cartridge before feeding to a Venusil MP C18 column (100 mm×2.1 mm, 3 μm).On the column, the sample solution was flushed with a 0.1% (v/v) formic acid and 5% (v/v) acetonitrile aqueous solution, as well as a 0.1% (v/v) formic acid and 95% (v/v) acetonitrile aqueous solution as the mobile phase at the flow rate of 0.3 mL·min-1 and the column temperature of 30℃.The multiple reaction monitoring mode was used for the qualitative and quantitative analyses, and the external standard method for the quantitative determination.When the concentration of clonidine hydrochloride or cyproheptadine hydrochloride was 1.0-100.0 μg·L-1, a linear correlation (r≥0.9995) existed.At the addition of 0.2, 2 and 10.0 μg·L-1 of the standard chemicals, the recovery rates were 83.5%-95.1% with an RSD (n=6) of 3.5%-8.7% and a quantity limit of 0.2 μg·L-1 (the signal to noise ratio >10).The newly developed method appeared to be precise, sensitive, accurate and simple to operate for the determination of clonidine hydrochloride and cyproheptadine hydrochloride contents in pig urine.
Spartina alterniflora from Min River Estuary Wetlands-A New Source for Forage

Abstract:
This study was to determine the commercial value and optimum harvest time of the common cordgrass, Spartina alterniflora, collected from the wetland of Min River estuaries in Fujian.The analytical results indicated that the grass' nutritional value peaked between June and September in a year.During this period of time, its crude protein reached 11.54%; calcium, 0.32%; phosphorus, 0.16%; NDF, 58.78%; ADF, 34.47%; ash, 7.93%; and NaCl, 3.35%.Consequently, S.alterniflora was found to be rich and complete in amino acids, and that it could be a potential source as forage.
Effect of Organic Cultivation on Yield and Quality of Flue-cured Tobacco

Abstract:
Effect of organic cultivation on the quality and commercial value of the flue-cured tobacco varieties, K326, KRK26 and HD, was studied.As compared with the conventional method, the organic cultivation caused significant tobacco yield decline (P<0.01) with an average reduction of 17.7%.On the other hand, its average market price increased 25.6%.The proportion of high-grade tobacco increased 19.2%, the proportion of medium-grade tobacco increased 9.8%, and the production value was not significantly different.The contents of TS and RS increased significantly (P<0.01); NIC, TN, K+ , main neutral aroma substances and total aroma substances decreased very significantly (P<0.01) or significantly (P<0.05).Sensory evaluation on the tobacco samples indicated that the organic tobacco leaves gave slight more alcoholic scent, slightly lower combustibility, less fragrant but more pleasant and delicate aroma, and less astringent nicotine flavor than its conventional counterpart, suggesting a possible somewhat superior commercial value.

Abstract:
In breeding a new variety of sweet corn for Fujian, various maize germplasms were introduced, screened and selected according to the specific climatic and ecological requirements for the region.Mintian 4, a cross between the parents, Mintianxi 138 and Mintianxi 186, displayed a high combining ability and desirable complementary results in crop characteristics.During the 2009-2010 regional field trials, it exhibited an average yield of 14 445.9 kg穐m-2 (i.e., an increase of 14.76% over CK), as well as the desirable properties on yield, quality, stability and adaptability.As a result, Mintian 4 had passed the provincial evaluation in 2012.

Teyou 5565 was bred by crossing the 3-line male sterile Longtepu A with the restorer line, Zhanghui 5565. The hybrid rice exhibited the desirable characteristics, including population uniformity, desirable color change upon ripening, high yield, good quality.The newly bred rive variety was certified by Fujian Crop Variety Committee (Zhangzhou) in 2012.
Optimization of Culture and Rapid Propagation for Phyllagathis fordii

Abstract:
Effects of hormones and activated carbon in medium on the proliferation and rooting of Phyllagathis fordii were studied for the culture optimization.Plant tissues from aseptic seedlings were used as the explants for the orthogonal design experiment [L9(34)].The result showed that (a) MS supplemented with 6-BA 1.2 mg·L-1+NAA 0.4 mg·L-1+KT 0.1 mg·L-1 was appropriate for the proliferation; (b) 1/2 MS supplemented with IBA 0.5 mg·L-1+activated carbon 1.0 g·L-1 was suitable for rooting; and (c) the survival rate were greater than 90% when the plantlets were transplanted to a soil medium containing 2/3 peat and 1/3 perlite.
Production, Nutrition and Safety of Young Shoots of Vegetable Milk Vetch

Abstract:
Field experiments and tests were carried out to evaluate the potential of growing Milk Vetch for food.The results showed that (a) the young shoot's yield decreased with plant types in the order of: medium flowering type>early flowering type>earlier flowing type; and (b) there were significant correlations between the shoot yield and plant growth stages or characteristics i.e., it descends as follow:freshyield>early flowering stage>plant height >stem diameter >budding stage).The young shoots contained more vitamin C, Cu, Mn and carbohydrate than pea sprouts, mung beans or water spinach.In particular, it had a high vitamin C content of up to 61.7 mg·hg-1.The contents of toxic and hazardous substances in the shoots were below the national non-public hazard vegetable safety standards.Thus, it appeared that the young shoots of Milk Vetch could be developed as an exotic edible vegetable.
Establishment of Tea Plantations with Improved Carbon Sequestration Ecosystem for Fujian

Abstract:
Examples of commonly found ecological problems of the tea plantations in Fujian included low surface coverage, severe soil erosion, low solar energy utilization in aging and young tea gardens, nitrogen over-fertilization, reduced carbon sequestration potential, and declined nutrient circulation and storage.To alleviate or minimize the shortfalls, a proposal for improvements is presented.A model was constructed aiming to increase the carbon sink and reduce the carbon emission.It was hoped that a high and stable yield, low-carbon tea production in the province could be resulted.The model includes designs of a tri-dimensional ecological tea garden and a compounded biological cycle tea garden, as well as the scientific fertilization and zero tillage.It could be estimated that the model of compounded and tri-dimensional ecological tea plantation would lead to consequences of increase in bio-carbon sequestration over 3.88?105 t and in soil organic carbon storage over 12.00?105 t annually if applied to 80% area of the 20?104 hm2 tea gardens in Fujian. Moreover,the amount of organic carbon storage would also be increased about 3.88?105 t by fertilizer optimization. With the establishment of technological model in tea plantations, the CO2 emission was equivalently reduced over 5.82?105 t, which significantly improved the carbon sequestration ability of tea gardens.

Based on the principles of agriculture ecology and sustainability, 6 key initiatives to aid the development in the province are proposed.They include (a) optimizing managerial design and improving developmental plans, (b) coordinating regional operations and specifying unit accountability, (c) generating technical models and encouraging innovative adaptations, (d) establishing system standards and providing developmental direction, (e) building industrial parks and fostering business transformation and upgrading, and (f) extending capital investment and augmenting economic growth.The implementation was expected to benefit energy conservation, pollution reduction, economic acceleration and productivity improvement for the province.

In the regions of China, there is a significant difference in the nature, crop growth and so on, and the production structure is diverse. The purpose of studying the tactics of regional accurate communication on agricultural science and technology is to explore a way how to adapt the contents of communication on agricultural science and technology to the local rural production practice. Abstract:
Industry-oriented leisure agriculture refers to leisure agriculture, which relied on local agriculture resources and tourism advantages, was stimulated by comprehensive agriculture technologies and tourism industry, and eventually became organic combination of both tourism and agriculture with special characteristics.In order to investigate the development model of industry-oriented leisure agriculture in West-strait Economic Zone, a case study was conducted in the eco-farm of Longtai Mountain in Fuzhou.By introducing kinds of superior cultivars of subtropical vegetables, fruits and flowers, advanced technologies and business management methods, a multi-functioned demonstration garden of leisure agriculture contained science education, tourism culture and leisure service was found, and significant operation efficiency and social ecological profits were achieved in this study.

Abstract:
Improving sorghum's disease resistance is one of the major objectives in our breeding program, as it is essential for the crop's high yield and desirable quality.In recent years, the advancement in molecular biotechnology fueled the significant progress in sorghum genome research.A large number of the critical resistance genes in sorghum have now been added on the genetic linkage map.It helps propel the studies on the physiology, resistance mechanism, resistance gene cloning, molecular marker assisted selection, favorable gene directed transfer and gene pyramiding on sorghum.In this article, notable achievements, such as the mapping of the QTL that resists drought and genes that resists diseases, pests, parasitic grasses or low temperatures, are summarized; the scientific challenges and anticipated future developments in the field of study are discussed; and specifically, some potential applications of the resistant gene mapping for sorghum breeding are presented.
