Abstract:
Shuangkangmingzhan has been proven to contain gene Gm6 through detection by molecular marker.It is the progeny of Kangwenqingzhan,which carries Gm6 that repels rice gall midge,crossed with Duoxi 1.It is the restored line against rice blast and rice gall midge resulting from years of natural and spontaneous selection process in multiple locations.The resistance test on Shuangkangmingzhan and its hybrid combinations with rice gall midge showed no significant difference between them.However,a significant difference existed between the restored line Minghui 100 and its hybrid combination Jinyouming100,which had no resistance resource.Shuangkangmingzhan is the restored line with restoring ability to the indica rice CMS line.And Teyoumingzhan is hybridized between Shuangkangmingzhan and Longtepu A.When the two were hybridized the resultant rice strain showed high grain yield and moderate resistance to rice blast.

Abstract:
Effects of nitrogen(N) and potassium(K) fertilizer application rates on the growth,development and yield of the hybrid rice in the field were observed.It was found that the tillering number,as well as the area and dry weight of the leaves increased with the increased rate of N and K fertilizer application.The fertilizer application rates exhibited a parabolic correlation with the crop growth rate,dry weight of stem and sheath,spike dry weight,total dry matter weight and grain yield.The hybrid rice was sensitive to N fertilizer.Therefore,the application rate of N is recommended for getting the highest grain yield,but K application for a balanced economic return.For every 100 kg grain in high yield cultivation,the optimum rates of application are 1.8 kg N and 1.6 kg K2O.

Abstract:
Effect of straw-returning amount and manner on the nutrient uptake,yield and quality of flue-cured tobacco were studied in field condition.The results showed:(1) Straw-returning treatment increased shoot biomass apparently,the decomposed straw and direct straw-returning treatments were better than straw-covering treatment.The effeciency of straw-returning was positively correlated to the straw-returning,but the effeciency of direct straw-returning treatment descended when the amounts excessed 5 250 kg·hm-2.(2)Sstraw-returning treatment improved the accumulation of N,P,K,Ca,Mg,Zn,Mn in plant,the decomposed straw and direct straw-returning treatments were superior to straw-covering treatment.The accumulation of N,P,K was positively correlated to straw-returning amount generally,but the correlation between the accumulation of Ca,Mg, Zn,Mn and the amounts was unapparent.(3)The straw-returning treatment decreased the amounts of reducing sugar,TN and RS/Nie in middle leaves,meanwhile,increased the amounts of Nie and TN in top and middle leaves,the effect on top leaves was most obvious by decomposed straw-returning way,the relationship between effect and amounts showed positive correlation,the effect on top leaves was most obvious by direct straw-returning way,between effect and amounts also showed positive correlation.(4)The treatments improved the economic characteristics of cured tobacco,but the effeciency treatments of decomposed straw-returning and direct straw-returning treatments decreased when over-retruning the straw amounts.

The application time,stage,insecticide and method are important factors in chemical control of Phyllotreta striolata(F).The lab and field toxicity tests showed that 48% EC of chlorpyrifos,40% EC of phoxim,80% EC of dichlorvos,20% EC of carbosulfan,24% SP of methomyl or 18% SL of bisultap could effectively control the pest in 48 hours.

The dynamic degradation and final residue of JS118 in cauliflower and soil were investigated in a Fuzhou suburb in 2005~2006.The results showed that(a) the 14-days average final residue in the cauliflower leaves was 0.20 mg·kg-1,(b) no residue detected in the soil in 2005,but 0.38 mg·kg-1 in 2006,and(c) 0.04 mg·kg-1 of the residue was found in the flowers.The half-life of JS118 in cauliflower leaves was determined to be 3.19 d in 2005 and 4.19 d in 2006.In the flowers,it was 4.07 d in 2006,and in the soil 6.43 d in 2005 and 5.73 d in 2006.

The relationship between the type of fruit-bearing branch and fruit position and the yield and quality of Cuiguan pear was studied.It was found that the total fruit yield on long or medium branches was more than that on short branches.The soluble solids content of the fruits born on long branches was higher than that in short branches,while the fruits form the medium branches had better shapes.Thus,proper pruning could improve the fruit yield and quality.Fruit position on a branch also significantly affected the yield and quality.The fruits on the first to the third positions tended to weigh more and have higher quality than the others.Especially,the second fruit on a branch had the highest yield,best shape and most soluble solids.Even though the weight of a single fruit positioned second did not significantly differ from the first and the third on a same branch,it would be prudent to keep the fruit born on the second position.

The effect of calcium on the growth of loquat seedlings was studied.The results indicated that loquat seedlings grew better when the concentration of calcium was 0.4 mmol·L-1.When the concentration was higher or lower,negative effects resulted.At calcium concentrations lower than 0.4 mmol·L-1,the plant height increased with the concentration increase.In the 11 th month the stem diameter was significantly correlated to the calcium concentration.But this correlation did not exist when the concentration was higher than 0.4 mmol·L-1.When the calcium concentration was below 0.4 mmol·L-1,the differences on fresh and dry plant weight,root volume,stem growth and diameter among treatments were significant.

To purify and rejuvenate the variety of C.reticulata Blanco "Fuju",pomelo was used as the pollen parent and Fuju the maternal plant to cultivate nucellus seedlings.Applying the top-grafting-exchanging-variety technique,the flowering and fruiting age of the nucellus seedlings was advanced by 2-3 years.The juvenile period of Fuju was cut short and the fruit quality improved while maintaining its original desirable characteristics.After years of observation and assessment on the top-grafting plants of nucellus seedlings,considerable quality variations were found among nucellus embryo plants.Three strains of superior varieties were identified: "Xinnong 1","Xinnong 2" and "Xinnong 3".They had high fruit yield,large fruit size,bright peel color,less seeds and high content of soluble solids.For example,Xinnong 1 strain contained 14.2% soluble solids,averaged 8 seeds,and weighed 118 g per fruit.This variety is recommended for expanded cultivation.

Based on the ORF7 gene sequence of the porcine reproductive and respiratory syndrome virus(PRRSV),a pair of primers were designed and synthesized.By means of the template of RNA of PRRSV,the RT-PCR technique for detecting PRRSV was established.The 443 bp specific fragments were obtained subsequently from our laboratory’s virocyte culture.Negative test results were found on the Porcine Circovirus Type 2,Pseudorabies Virus,Classical Swine Fever Virus,and Marc-145 cells.The amplified sequence of PCR product was 92%-98% in agreement with those published data on PRRSV strains.The sensitivity of RT-PCR reached 2.39TCID50.In 2004-2005,23.4% of the 81 clinical specimens tested to be positive.The results showed that the newly developed RT-PCR method had high specificity and sensitivity,and that it could be applied for PRRSV diagnosis and epidemiological investigation.

A virus strain,E05,was isolated from oviduct mucosa of breeder Muscovy duck,which showed irregularity and reduction in egg-laying.The virus was identified as an egg-laying reduction syndrome virus(EDSV) by haemagglutination and haemagglutination inhibition tests.According to the conserved region of duck original EDSV sequence,a pair of primers was designed for PCR.The product of the PCR was ligated with T vector and introduced into competent cells.Positive recombinant was selected for sequencing.The sequence of PCR product was compared with that of the 100 kD gene found in GenBank nucleotide database.The result showed that the amplified product from the isolated virus was completely identical with the counterpart of the EDSV.

Chromosome specimen of kidney cells was prepared by using colchicine-low osmosis-air drying and Sumner’s(1972) C-banding staining method.The karyotype and C banding of Trichogaster trichopterus sumatranus were observed.The results showed that the karyotype formula of Trichogaster trichopterus sumatranus was 2n=46 t,NF=46,the lightly and darkly stained C-bands existed on centrometric and terminal spots of most chromosomes,and there was no heteromorphic sex chromosomes.Their relationship and systematic evolution were discussed by comparing with several other species in the same family.

A pair of specific primers of gene encoding phenol hydroxylase was designed by oligonucleotide high conservative sequence.A 684 bp fragment was amplified from Alcaligenes faecalis BC2001’s total DNA.After sequencing and alignment by Blast,the sequence share 97% homology with phenol-degrading Proteobacterium L-18 strain(accession number:AY346142).Another phenol-degrading experiment showed that pH 6 was optimum for the growth of BC2001.It exemplified the BC2001 strain showing its phenol-degrading ability at 300 mg·L-1 of phenol concentration.

Cell biomass and pH of Geobacillus stearothermophilus CHB1 in culture medium were used as indices in the optimization study.The results indicated that different carbon and nitrogen sources significantly affected the microbial growth.By using orthogonal experimentation design,the optimal medium composition was obtained: 0.5% beef extract,0.9% soy peptone,0.2% NaCl,0.1% K2HPO4 and 0.075% KH2PO4.It was found that the biomass resulted from the newly formulated medium was ten times greater than that from the standard fermentation medium.

The current status of the agricultural soil resource in Fujian is presented in this article.The problems facing the provincial agricultural soil,including serious soil erosion,deterioration of fertility,low fertilizer efficiency and severe pollution,are analyzed.Based on the information and analysis,critical control measures to improve soil quality and curtail further deterioration are recommended.The applicable technologies included: surveying,monitoring and evaluating soil quality,developing long term and fixed-site soil quality tests,recovering ecosystem and restoring soil conditions,implementing fertilizer efficiency improvement measures,coupling water and fertilizer management,etc.

An external method based on liquid chromatograph-electrospray ionization-mass spectrometry was developed for the determination of low-level residues of four nitrofuran in eel muscle.The procedure entailed an acid-catalyzed release of protein-bound metabolites,followed by the derivation,liquid-liquid extraction,positive electrospray ionization applying multiple reaction and monitoring two transitions for each compound.The standard curve was reliable,the LOQ was 0.5 ng·g-1,the nitrofuran metabolite recovery rates at 5 fortification levels(i.e.,0.5,1.0,2.0,5.0,10.0 ng·g-1) were 70%-130%.The LOD was 0.25 ng·g-1.The method was rapid,simple economical and its LOQ below the minimum performance limit of 1 ng·g-1 set by Japan and the EU.

A method for rapid separating and determining γ-amino butytic acid(GABA) in plant products by using the HITACHI L-8800 amino acid analyzer was developed.The time required for the new procedure was 30 min in comparison to 53 min for the standard analysis.The determination results using the two methods were similar.At GABA concentration of 100 nmol·ml-1,the coefficient of variation of the peak potential and peak area was 0.17% and 0.23%,respectively.The recovery rates of GABA with the new procedures was 97.89%-101.2%.The relative errors of the determination in brown rice and traditional Chinese medicines by using the two methods were less than 3%.It was thus concluded that this modified method was stable,rapid,accurate and suitable for the determination of γ-amino butytic acid.

Based on the facing situation in rural infrastructure construction after the reform of rural taxes and administrative charges,the chance and challange were analyzed;the achievement and problems existing before and after the reform were pointed out;the basic consideration on infrastructure construction in post-period of the reform of rural texes and administrative changes were put formard,that is,taking a scientific concert in development,concerning with masses intervests,improving the facilitates of the rural traffic,information,education,energy,provision,drinking warter,flood control,disaster prevention through above analysis,the countermeasures to fasten the progress of rural infrastrueture construction were disccused in order to make a beneficial recycle between investment and profit in the rural infrastructive construction.

Abstract:
Vegetables accumulate nitrate residues easier than other crops.Thus,studying the factors affecting the accumulation in vegetables and possible control measures has been the focus for research scientists.This paper gives the current status on the study summarizing the internal factors,such as species,variety and accumulation site,as well as,the external factors,such as,fertilization,illumination,moisture and harvest stage.These factors affect the absorption and dynamic change of nitrate in the vegetables.Control measures,such as,by improving the agronomic practices,regulating environmental conditions and breeding new vegetable varieties,in order to reduce the nitrate pollution are presented.

Using the polyethylene glycol(PEG) simulated drought stress test,the growth,leaf and stem relative water content(RWC) and protoplasmic membrane penetration (RPP) of Arachis pintoi cv.Amarillo and A.duranensis krapov.& W.C.Greg were studied with 0%,5%,15% and 25% PEG for 24 hours.It was found that the PEG drought stress restrained the growth and physiology metabolism of A.pintoi and A.duranensis to some extent.The effect on the leaves was more significant than the stems.A.pintoi showed a slightly stronger drought resistance than A.duranensis,but not significantly.The regression analysis indicated that,under the stress test,significant(P0.05) negative linear correlations existed between the drought condition and the RWC and RPP of A.pintoi’s stems and A.duranensis’ leaves and stems.This indicated that a synergism might present between RWC and RPP of A.pintoi and A.duranensis.

Controlling the severely spreading Eichhornia crassipes has been a top priority project for the Shuikou Reservoir in Fujian.This article describes the situation prior to the implementation of the "Land-Water Mechanical Salvaging Technique," objectives set forth for the project,methodology development and detailed equipment requirement and operation procedures of the control technique.It is hoped that by promoting the application of this newly developed method,the invasive overgrowth of Eichhornia crassipes in the reservoir,lake,river,inland water-network could finally be controlled.
