Isolation and Preliminary Identification of Avian Adenovirus Serotype 4

摘要: 从疑似心包积液综合征的病死鸡组织中分离获得1株病毒,该病毒无血凝活性,提取其核酸经禽流感病毒、新城疫病毒、鸡传染性支气管炎病毒、禽脑脊髓炎病毒、禽白血病病毒、传染性法氏囊病毒、禽偏肺病毒和包涵体肝炎病毒特异引物扩增均为阴性,而经禽腺病毒4型特异性引物检测为阳性,则将其暂命名为禽腺病毒4型FJ1674株。将其扩增产物回收后克隆,经序列测定分析表明该株病毒与国内外禽腺病毒4型毒株的同源率为99.4%~100%;经遗传进化分析表明,其与禽腺病毒4型关系密切,共处同一分支。Abstract: A viral strain, designated as FJ1674, was isolated from dead chickens that showed hydropericardium syndrome. The gene fragment of the strain, which lacked the hemagglutination activity, could be amplified by the specific primers of avian adenovirus serotype 4 against those of the viruses of avian influenza, newcastle disease, avian infectious bronchitis, avian encephalomyelitis, avian leukemia, infectious bursa, avian metapneumo, and inclusion body hepatitis. Thus, FJ1674 was believed to belong to avian adenovirus serotype 4.The sequencing analysis revealed its high homology (99.4%-100%) between FJ1674 and its counterpart in GenBank. The phylogenetic analysis also showed that FJ1674 was close to viruses of the same serotype, and could be subdivided into the same isolate cluster.
