Studying on Somatic Embryo Induction of Fortunella japonica

摘要: 采用金桔无菌苗叶片为外植体, 在单因素试验的基础上, 通过正交试验研究不同生长调节剂对金桔胚性愈伤组织诱导的影响。结果表明:MS+2, 4-D 2.0mg·L-1+BA 0.5mg·L-1处理最适宜金桔胚性愈伤组织诱导, 其中淡黄色愈伤组织团块胚胎发生频率较高;在附加BA 0.05mg·L-1的MS培养基上淡黄色愈伤组织块可形成胚状体;胚状体试管苗在1/2MS+NAA0.1mg·L-1培养基上, 生根率可达80%, 移栽成活率达70%Abstract: Based on single factorial experiment, the sterile seedlings of Fortunella japonica were used as explants for studying the effects of various growth regulators on inducing embryogeny with orthogonal test.The results showed:the MS+2, 4-D 2.0mg·L-1+BA 0.5mg·L-1 was the best culture medium for inducing embryonic callus.Sucrose at MS culture medium with BA 0.05mg·L-1 gave a maximum embryogenesis from the light yellow callus tissue of leaf.Planting somatic embryos on 1/2MS culture medium with NAA0.1mg·L-1, the rooting rate was 80%, the survive rate was 70%.
