Application of PCR-DGGE on Analyzing Microbial Diversity in Intelligent Biogas Digester

摘要: 采用变性梯度凝胶电泳(PCR-DGGE)技术研究智能化上流式沼气池微生物群落结构的动态变化。结果表明:沼气池中微生物种类丰富,并且拥有优势种群,主要有:稳杆菌属、热单细胞属、杜檊氏菌属、戈登氏菌属、芽孢杆菌属、拟杆菌门、变形菌门的α类群和厌氧细菌门。各细菌种类不同发酵层、不同发酵时间对微生物多样性存在影响。DGGE图谱中的优势条带16SrDNA基因序列经比对后都属于不可培养细菌。说明利用PCR-DGGE技术研究智能化沼气池中微生物多样性是可行的。Abstract: The bacterial community and diversity were revealed by using PCR-DGGE technique,which were associated with biogas slurry of up-flow intelligent biogas digester.The results showed that the majority genus of bacteria were Empedobacter sp.,Thermomonas sp.,Duganellasp.,Gordoniasp.,Bacillus sp.,Bacteroidetes bacteriumand anaerobic bacterium.The sequences of 16SrDNA predominant bands in DGGE were compared with NCBI database.The results showed that the bacterial community was abundant in up-flow intelligent biogas digester,but its community structure was changed variously with fermentation time phases and fermentation layer phases in intelligent biogas digester.The most of predominant bacterial genus were non-cultural bacteria.The results suggest that the DGGE technique is reliable in applying to analyze microbial diversity in intelligent biogas digester.
