Extraction and Deproteinization Technology for Crude Polysaccharides in Radix Pseudostellariae

摘要: 太子参多糖是太子参的有效成分之一。对太子参中粗多糖的热水浴加热提取工艺进行研究,对料液比、提取温度、提取时间、提取次数分别进行单因素试验和L9(34)正交试验,结果表明最佳工艺料液比1∶20,温度100℃,回流提取90min,提取3次。用Sevage法、三氯乙酸法、三氯乙酸-正丁醇法3种方法对太子参粗多糖去除蛋白处理效果进行比较,结果显示三氯乙酸-正丁醇法的蛋白质去除效果较好,其多糖纯度为81.2%,蛋白含量为9.93%,蛋白去除率为60.14%。Abstract: Polysaccharide is one of the active ingredients extracted fromradix pseudostellariae.In this study,the solid-liquid ratio,temperature,extracting duration and times in crude polysaccharide extraction of radix pseudostellariae were investigated by hot water bath,using both the single-factor test and L9(34)orthogonal test.The results showed that the optimum parameters for extraction were with the ratio of solid to liquid 1∶20,extracting temperature 100℃,reflux extraction time 90min and extraction for 3times.The deproteinization effects were compared among three methods,the Sevag reagent,trichloroacetic acid,and trichloroacetic acid-butyl alcohol,and the results presented that trichloroacetic acid-butyl alcohol was the best for deproteinization,with 81.2% crued polysacchsrides and 9.93% protein,indicating that the deproteinization rate was 60.14%.
