Optimization for Regeneration System of Embryonic Tips of Vegetable Soybean

摘要: 以福建省菜用大豆新品种闽豆5号的胚尖为材料,研究次氯酸钠消毒方法及不同浓度潮霉素B培养基对不定芽的诱导和生根的影响。结果表明:以10%的次氯酸钠消毒10min为宜;芽伸长阶段潮霉素B筛选压力为7mg.L-1;生根培养基不添加6-BA效果更佳,即1/2 MS盐+1/2B5有机+3%蔗糖+0.56%琼脂+1.0mg.L-1 IBA,pH 5.8。Abstract: Embryonic tips from 'Mindou No.5' were used in testing the effect of NaClO on sterilization and homomycin B on screening stress for inducing of adventitious buds and root growing.The result showed that the optimal effect on sterilization was 10% NaClO for 10 min;the screening stress of hygromycin B was 7 mg·L-1 at elongation stage of buds from embryonic tips.Comparing culture mediums,the rooting culture medium presented better in root growing without 6-BA,the medium consisting of 1/2 MS salt+1/2 B5 organic matter +3% sucrose+0.56% agar+1.0 mg·L-1 IBA,pH 5.8.
