Transgenic Rice with mgfp4 Gene Mediated by Agrobacterium tumefaciens

摘要: 通过农杆菌介导法将携带mgfp4基因的植物表达载体pCAGFPHyg导入水稻明恢86中,获得101个独立克隆的转基因植株。PCR检测和Southern blot分析证明外源基因已整合进水稻基因组中,T0代种子的分离分析结果表明大部分转基因植株为单T-DNA插入。荧光检测及RT-PCR分析结果显示mgfp4基因能在水稻中表达,但其发出的绿色荧光检测效果不甚理想。Abstract: Plant expression vector,pCAGFPHyg,carrying mgfp4 gene was introduced into rice Minghui 86(Indica Sativa L.) mediated by Agrobacterium tumefaciens.The transgenic rice containing 101 independent clones was obtained.PCR and the southern blot analysis ensured that the exogenous genes were indeed integrated into the rice genome.The result of T1 segregation analysis from T0 rice seeds indicated that most of the transgenic plants were inserted by a single T-DNA.It was confirmed by the fluorescence microscopic and RT-PCR detections that the mgfp4 gene was expressed in the transgenic rice,although no acceptable result of green fluorescence was secured.
