Identification and Preliminary Analysis of a Male Gametophytic Mutant mga36in Rice

摘要: 利用Tail-PCR分离1个T-DNA插入导致的水稻配子体不育类型的雄配子不育突变体mga36的侧翼序列。比对结果显示T-DNA插入在水稻第7号染色体上1个未知蛋白基因的内含子区。RT-PCR表达分析发现候选基因MGA36在花粉成熟期的根、茎、叶、小穗中均有表达。扫描电镜观察发现突变体约50%花粉略微凹陷,表明突变发生在花粉发育晚期。综合结果显示候选基因MGA36在水稻花粉发育晚期起重要作用。Abstract: An identified male gametophytic sterile mutant mga36 (Male sterility abortion 36) in rice (Oryza Sativa L.) was resulted with the insertion of a T-DNA. This special sequence flanking T-DNA was isolated with Tail-PCR technique. Blast search result showed that the insertion of T-DNA located in the intron of an unknown protein gene in chromosome 7. RT-PCR expression analysis revealed that, in pollen mature stage, the candidate gene MGA36 had expressed in all test tissues including roots, stems, leaves and spikes. The observation of scanning electron microscope (SEM) for mature pollens with mutant mga36 indicated that 50% the pollens were slightly concave. The study suggested that the gene MGA36 played an important role in the late stage of rice pollen development.
