甘蔗染色体及Giemsa-C带研究初报
A Preliminary Study on Giemsa C-Banding of Sugarcane Chromosomes

摘要: 本文研究了甘蔗属三个种4份材料染色体的Giemsa-C带。结果表明:(1)甘蔗根尖经过预处理→固光→酶解→涂片→熟化→BSG分带处理程序,可以诱导出染色体的GiemsaC带,长时间熟化处理是取得分带成功的关键。(2)蔗茎段经过变温培养,诱导幼根,可以促使根尖细胞同步分裂,提高中期分裂相的比例。(3)二个刈手密野生种和热带种异染色质程度较低,是进化上较为原始的种;中国种异染色质含量高是较为进化的种,并对中国种的起源进行了讨论。研究结果可为甘蔗遗传育种和细胞学研究提供参考。Abstract: Giemsa C-banding of the chromosomes of four clones of three sugarcane species,S.spontaneum,S.sinense and S officinarum,were studied in the paper.The results are showed as follows.(1)After the root tip of sugarcane experienced the treating procedure: pretreatment→fixa-tion→enzymatic isolization→smear→maturation in dry air→BSG banding treatment,the Giernsa C-banded chromosome would be induced;and the time of maturing treatment in dry air is the key of success,(2)The two species of S,spontaneum and S.officinarum had low content of heterochro-matin and they were more original species in sugarcane evolution.S.sinense had higher heterochro-matin content and so it is more developed species.The origin of S.sinense was also discussed in the paper.
