菲律宾引种香蕉花叶病毒的鉴定
Etiological Identification of Mosaic Disease in Banana Imported from the Philippines

摘要: 根据病毒生物学特性、粒体形态和血清学性质,明确在厦门地区种植的菲律宾引种香蕉(Musa sp.)表现花叶症状病株上分离的一个病毒分离物是属于黄瓜花叶病毒(Cucumher mosaic cucumovirus,CMV)亚组Ⅱ。该分离物能通过摩擦接种侵染供试11科52种(或品种)植物中的10科47种(或品种);能够由棉蚜(Aphis gossypii)传播;失毒温度为50~55℃,稀释终点为10-3~10-4,体外存活期为24~48h.提纯病毒粒体为球状,直径约28~30nm.用该分离物提纯病毒免疫家兔制备的抗血清,经琼脂双扩散方法测定效价为1:64.用法国提供的CMV单克隆抗体,经双抗体夹心ELISA(DAS-ELISA)测定,该分离物与ToRS血清组(亚组Ⅱ)的单克隆抗体具有强阳性反应。Abstract: One virus isolated from mosaic disease of banana introduced from the PhilippInes and grown in Xiamen area,was identified as an isolate of cucumber mosaic cucumovirus subgroup Ⅱ,based on its bio-logical propertles,particle morphology and serology of the virus isolate. The virus isolate could infect 47 species (or varieties) of ten famllies from 63 species (or varieties) of 11 tested families and be also transmitted by Aphis gossypii. The thermal inactivation point for the virus isolate was 50-55℃;the dilution end point was 1:10-3-10-4, and the longevity in vitro was 24-48h. The electron microscopic examination of the purified virus particle preparations showed isometric particles of ahout 28-30nm in diameter.The antiserum obtained had a homologous titre of 1:64 in gel diffusion tests. The virus isolate had strong positive reaction to monoclonal antibodies of CMV ToRS serogroup in double antibodies sandwich enzyme-linked immunosorbent assay.
